Isoenzyme Analysis
Isoenzyme analysis is generally requested but if the cell line is derived from a heterohybridoma (e.g. human/mouse), DNA fingerprinting is sometimes required. The analysis provides a permanent record of the cell line isoenzyme profile, which is unique to each animal species.
Standard isoenzyme analyses included determination of the isoenzyme gel electrophoresis banding pattern for at least three enzymes. This protocol is intended to verify cell line identity and to detect cell line contamination by comparing the migration distances of 3-4 different isoenzymes chosen from the of the following:
- Glucose-6-Phosphate Dehydrogenase (G6PD)
- Lactate Dehydrogenase (LD)
- Malate Dehydrogenase (MD)
- Nucleoside Phosphorylase (NP)
- Aspartate Aminotransferase (AST)
- Peptidase B (PepB)
- G-Protein Coupled Receptors (GPCR)
- cAMP Phosphodiesterase Assay
- Protein Kinase Assay (PK)
