Glycoprotein and Oligosaccharide Analysis
Glycoprotein detection: After SDS-PAGE and Western blotting of glycoproteins, immobilized proteins are oxidized to generate free aldehyde groups for incorporating biotin hydrazide into the molecule. Then the glycoprotein complex is detected by streptavidin-alkaline phosphatase and NBT/BCIP color development reagents.
Oligosaccharide Analysis: Profiling of N- and O-linked oligosaccharides from glycoproteins is achieved by enzymatic treatment of glycoprotein, followed by fluorescent labeling and electrophoretic separation of labeled oligosaccharide.
